Destabilized mitochondria generate mitoplasts, which affect organelle turnover.

  • An efficient cellular surveillance system regulates the turnover of endoplasmic reticulum (ER) and mitochondria under stress.
  • AMFR, an ER-associated E3 ligase, degrades outer mitochondrial membrane proteins and triggers mitophagy.
  • High levels of AMFR in stressed mitochondria lead to the formation of mitoplasts, bringing the inner mitochondrial membrane closer to the ER.
  • RETREG1 interacts with OPA1 to aid in the formation of the mitophagophore, contributing to the degradation of inner mitochondrial membrane proteins.
  • Both OPA1 and inner mitochondrial membrane proteins undergo RETREG1-dependent autophagosomal degradation, differing from most outer mitochondrial membrane proteins.
