Abnormalities in mitochondrial processes are associated with numerous neurodegenerative diseases.

  • Mitochondria are essential for ATP generation, calcium balance, and reactive oxygen species regulation in neurons.
  • Mitochondrial dynamics, including fission, fusion, and mitophagy, are critical for maintaining neuronal health.
  • ROCK1 promotes excessive mitochondrial fission by adding phosphate groups to Drp1, contributing to neuronal cell death.
  • ROCK2 inhibits mitophagy by blocking the PTEN protein, leading to reduced mitochondrial quality control.
  • Impaired mitophagy can result in oxidative stress and degeneration of neuronal cells, which is a hallmark of neurodegenerative diseases.
  • Targeting ROCK isoforms may offer therapeutic potential by improving mitochondrial dynamics and neuroprotection in various neurodegenerative diseases.
