RNA Interactions Shape the Structure and Behavior of Lipid Nanoparticles

Updated

Abstract

A 46% slowdown in frequency fluctuation dynamics is observed in RNA-loaded lipid nanoparticles compared to empty ones.

  • Lipid nanoparticles (LNPs) exhibit pH-dependent structural changes that affect their delivery performance.
  • At pH 4, increased electrostatic interactions between RNA and protonated MC3 result in broader spectral features, indicating greater interfacial heterogeneity and hydration.
  • Raising the pH from 4 to 8 narrows the ester carbonyl band by approximately 10 cm-1 and leads to similar dynamics for both loaded and empty LNPs.
  • Molecular dynamics simulations suggest that MC3 protonation influences RNA binding, interfacial hydration, and lipid packing.
  • Electrostatic interactions between RNA and lipids may play a critical role in determining the local environment within LNPs.
