RNA Size and Shape Affect the Measured Acidity of Ionizable Lipid Nanoparticles

Abstract

RNA payload size impacted the apparent pKa of lipid nanoparticles (LNPs) by 0.1-0.5 pH units.

  • Different ionizable lipidoids were used to encapsulate RNA cargos ranging from 21 nucleotides to 4.5 kilobases.
  • The structure of the ionizable lipidoid influenced the direction and magnitude of changes in LNP pKa.
  • Short RNA sequences lowered the apparent pKa of LNPs with the monoamine lipidoid SM-102 compared to mRNA.
  • For multiamine lipidoids cKK-E12 and XMAN6, longer RNA sequences led to a consistent decrease in apparent pKa, plateauing at approximately 1.9 kilobases.
  • A pronounced difference in apparent pKa values was observed with the triamine lipidoid XMAN6, indicating the significance of amino group multiplicity.
