Using Natural Plant Compounds to Influence Mitochondrial Splitting and Recycling Processes in Brain Disorders by Targeting ROCK Proteins

Updated

Abstract

A library of 1,223 natural product compounds was screened for their effects on mitochondrial dynamics related to neuroprotection.

  • Mitochondrial dysfunction, characterized by excessive fission and impaired removal of damaged mitochondria, is linked to neuroinflammation and neurodegenerative diseases.
  • Rho-associated coiled-coil kinase isoforms (ROCK1 and ROCK2) are identified as key regulators of mitochondrial processes, with ROCK1 promoting fission and ROCK2 inhibiting mitophagy.
  • Overexpression of ROCK isoforms leads to increased reactive oxygen species (ROS) production and decreased ATP generation, which can compromise neuronal health.
  • Natural phytochemicals derived from Indian medicinal plants were screened for their potential to inhibit ROCK isoforms, offering a safer alternative to synthetic inhibitors.
  • Scopolin and Lycorine were identified as promising ROCK1 inhibitors, while Cytisoside and N-Methyltyramine showed strong inhibition of ROCK2.
  • Coumarin demonstrated stable interactions with both ROCK1 and ROCK2, suggesting its dual inhibitory potential and favorable properties for blood-brain barrier permeability.
