Rosmarinic acid may reduce heart damage and cell aging caused by doxorubicin by affecting the 14-3-3/Foxo1 signaling pathway

Abstract

Rosmarinic Acid (RA) significantly improved survival rates and cardiac function in a murine model of doxorubicin-induced cardiotoxicity.

  • RA decreased senescence markers in cardiomyocytes, as evidenced by reduced SA-β-gal activity and downregulation of p16/p21.
  • RA treatment was associated with diminished secretion of inflammatory factors linked to cellular aging, such as IL-6, IL-1β, and TNF-α.
  • Mechanistically, RA increased the expression of 14-3-3θ and enhanced the phosphorylation of Foxo1, inhibiting its movement into the cell nucleus.
  • In the mouse model, RA reduced myocardial atrophy, fibrotic remodeling, and mitochondrial damage, indicating protective effects against cardiac deterioration.
