Safety and immune response of the mRNA mpox vaccine BNT166a in a first human trial with increasing doses

Essence

BNT166a had an acceptable early safety profile and induced mpox-directed antibody responses in healthy adults.

Evidence

This open-label, uncontrolled, non-randomized phase 1 dose-escalation trial gave two BNT166a doses to 48 VACV-naive and 16 VACV-experienced participants.

Caveat

The trial was small and uncontrolled, measured antibody responses rather than mpox protection, and neutralizing responses waned over 12 months.
