Safety and immune response of the rVSVΔG-ZEBOV-GP Ebola vaccine in healthy adults in a controlled dose study

Abstract

A geometric mean IgG ELISA endpoint titre of 1624 was observed at the 2 × 10PFU dose on day 28.

  • The vaccine was well tolerated, with most adverse events being mild to moderate and occurring within the first day post-vaccination.
  • At the 2 × 10PFU dose, arm pain and local tenderness were significantly more common in vaccine recipients compared to placebo.
  • Self-limited post-vaccination arthritis occurred in 4.5% of vaccinees, with no clear dose relationship.
  • Robust antibody responses were evident in most participants by day 14, with seroconversion rates of 95.7%.
  • IgG and neutralizing antibody titres increased with higher vaccine doses, indicating a dose-response relationship.
  • Immunological responses were sustained for up to one year after vaccination.
