Safety, side effects, and immune response of the self-amplifying mRNA COVID-19 booster vaccine GRT-R910 in healthy adults

Abstract

Forty-eight participants were enrolled in a phase 1 study evaluating the safety and immunogenicity of the GRT-R910 vaccine.

  • Mild-to-moderate systemic reactions occurred in most participants, with 17% experiencing severe reactions.
  • Neutralizing antibody responses increased significantly after vaccination, with geometric mean fold rise (GMFR) values varying by age and dose.
  • Participants aged ≤60 years showed GMFR values of 5.2 and 6.1 at Day 29 and Day 181 after receiving a 3 μg dose, respectively.
  • In those aged >60 years, GMFR values reached 8.1 and 8.2 after a 3 μg dose, but were lower for higher doses.
  • The persistence of GMFR responses until Day 181 suggests that the vaccine boosts sustained antibody levels.
