Schaftoside from fenugreek may reduce depression and memory problems through the gut-brain connection in stressed mice

Updated

Abstract

Schaftoside, at doses of 20, 50, and 125 mg kg-1, significantly improved depressive-like behaviors in a chronic restraint stress mouse model.

  • Schaftoside reduced hippocampal neuronal damage and neuroinflammation, restoring levels of serotonin, norepinephrine, and acetylcholine.
  • Improvement in intestinal barrier integrity was observed, indicated by enhanced protein expression of tight junctions and reduced pro-inflammatory cytokines.
  • Schaftoside altered gut microbiota composition by decreasing stress-associated Bacteroides and increasing butyrate-producing Butyricicoccus.
  • Metabolomics analysis showed restoration of tryptophan and alpha-linolenic acid metabolism pathways.
  • Significant correlations were found between specific gut genera, fecal metabolites, and markers of inflammation and neurochemistry.
  • Fecal microbiota transplantation from schaftoside-treated mice replicated behavioral and anti-inflammatory effects in recipient mice.
