Abstract

The compound (+)-EU1180-453 doubles the NMDAR response at lower concentrations compared to its predecessor (+)-CIQ.

  • (+)-EU1180-453 is a selective modulator of GluN2C and GluN2D subunits in NMDARs.
  • It enhances glutamate efficacy by prolonging deactivation time and increasing peak response amplitude.
  • (+)-EU1180-453 selectively boosts synaptic NMDAR-mediated charge transfer in hippocampal interneurons but does not affect CA1 pyramidal cells.
  • This potentiation enhances inhibitory output from GABAergic interneurons onto CA1 pyramidal cells in a GluN2D-dependent manner.
  • The compound shifts excitatory-to-inhibitory coupling towards increased inhibition and enhances gamma-band power in hippocampal slices.
