Self-amplifying mRNA vaccine combining multiple targets against Staphylococcus aureus using computer-based design and structure modeling

Abstract

A bivalent self-amplifying mRNA vaccine cocktail targeting the ClfA, Hly, and SraP antigens of Staphylococcus aureus was developed.

  • No licensed vaccine currently exists for preventing invasive S. aureus infections in humans.
  • The candidate vaccines, SaBVax807 and SaTVax876, were designed using a combination of computational methods to identify key immunogenic regions.
  • Molecular simulations validated the binding interactions of the vaccine constructs with antibodies and human leukocyte antigen alleles.
  • Population coverage analysis for SaTVax876 indicated its potential applicability across sixteen geographically diverse regions.
  • Safety and immunogenic potential were rigorously evaluated, showing favorable profiles for both candidate vaccines.
