A 3D Model of Aging Lung Cell Clusters to Study Cell Aging and Fibrosis Signals

Abstract

A novel alveolosphere model was established to study epithelial senescence related to idiopathic pulmonary fibrosis (IPF).

  • The model was created by culturing alveolar epithelial cells with or without basement membrane components.
  • Cell viability assessments showed no cytotoxicity at 3, 6, and 8 days of culture.
  • Expression of specific markers indicated the maintenance of key epithelial and alveolar characteristics.
  • Induction of cellular senescence with doxorubicin led to increased levels of senescence-associated and pro-fibrotic markers.
  • The findings indicate that this alveolosphere platform could facilitate investigations into the role of epithelial senescence in IPF pathogenesis.
