Treatments targeting aging cells improve tumor control and survival in mouse cancer models

Abstract

Among 36 reviewed studies, senolytic drug co-treatment consistently reduced tumor burden compared to senogenic treatment alone.

  • Senolytic and senogenic combination therapy was evaluated across various cancer types, including colorectal, breast, and lung cancers.
  • Most combination therapy groups showed decreased levels of senescence markers like SA-β-gal, p21, p53, and p16, indicating reduced cellular aging effects.
  • The SASP cytokine IL-6 was notably suppressed in most models, suggesting modulation of the inflammatory response associated with senescence.
  • Reduction in Ki-67 levels and increased Caspase-3 activation were observed, indicating decreased cell proliferation and enhanced apoptosis.
  • In several studies, downregulation of BCL2 and elevation of γ-H2AX were noted, further suggesting increased apoptotic activity and DNA damage.
