Could drugs targeting aged cells help treat giant cell arteritis?

Updated

Abstract

Cellular senescence is implicated in the pathophysiology of giant cell arteritis (GCA), particularly through the accumulation of senescent cells in affected arteries.

  • Senescent cells in GCA produce pro-inflammatory substances that may contribute to ongoing inflammation and tissue damage.
  • Senotherapeutics, which target senescent cells, are classified into senolytics that kill these cells and senomorphics that suppress their harmful effects.
  • Potential senolytics include fisetin, quercetin, curcumin, and dasatinib, which also have anti-inflammatory and vascular-protective properties.
  • Senomorphics such as resveratrol and metformin may help alleviate vascular inflammation and restore immune balance.
  • Current treatments for GCA primarily focus on reducing inflammation and may not effectively address age-related factors that lead to vascular injury.
