Psychedelics may transiently relax overly rigid cortical priors and recalibrate dysfunctional neural circuits in Autism Spectrum Disorder (ASD).

  • Dysregulated serotonergic signaling and impaired neuroplasticity are implicated in the pathophysiology of ASD.
  • Psychedelics such as psilocybin and LSD act as high-affinity 5-HT receptor agonists and may enhance synaptic plasticity.
  • These compounds could modulate cortical oscillations and suppress neuroinflammation by shifting microglial phenotypes.
  • Preclinical and early human studies indicate potential improvements in sociability, sensory responsiveness, and behavioral flexibility.
  • There is an urgent need for rigorous clinical trials to establish safety and efficacy of psychedelics in treating ASD.
