Boosting serotonin in three ways helps prevent sudden death from seizures in Dravet syndrome mice

Abstract

Fluoxetine at 30 mg/kg and fenfluramine at 1-10 mg/kg significantly reduced or eliminated spontaneous seizure-induced mortality in Dravet mice.

  • Dravet mice exhibited spontaneous seizures that progressed from wild running to tonic seizures, sometimes leading to sudden death.
  • Fluoxetine reduced the incidence of spontaneous seizure-induced mortality, but only at a dosage of 30 mg/kg.
  • Fenfluramine provided full protection against spontaneous seizure-induced mortality at doses between 1-10 mg/kg.
  • SR 57227 significantly lowered spontaneous seizure-induced mortality at a dose of 20 mg/kg, but not at lower doses.
  • The effects of these treatments on spontaneous seizure-induced mortality were consistent across both male and female mice.
