How sex and APOE4 gene affect blood vessel amyloid buildup in Alzheimer's disease

Abstract

Overall CAA scores were higher in males than females in a study of 428 autopsied brain samples from Alzheimer's disease cases.

  • Carrying one or more APOE4 alleles is associated with higher overall CAA scores.
  • Biochemical analysis shows significantly elevated levels of detergent-soluble and detergent-insoluble Aβ40, and insoluble apoE in individuals with severe CAA or APOE4.
  • The ratio of Aβ40/Aβ42 in insoluble fractions increases in the presence of CAA or APOE4 but is negatively associated with male sex.
  • Levels of insoluble Aβ40 are positively associated with levels of insoluble apoE, influenced by CAA status.
  • Levels of insoluble Aβ42 correlate with apoE levels regardless of CAA status.
