Single-cell analysis reveals aging-related changes and control networks in blood-forming bone marrow cells

Abstract

A novel hematopoietic subpopulation was identified in 3.19% of aged bone marrow samples.

  • The identified subpopulation activates the cellular senescence pathway and enhances inflammatory interactions with CD8⁺ T cells.
  • Key signaling modules, including CAPN1, MAP2K1, and JUND, were highlighted through multi-omics analyses.
  • Cox regression modeling indicated that a module based on CAPN1, MAP2K1, and JUND has predictive value for aging.
  • In two independent cohorts, this module demonstrated area under the curve (AUC) values of 0.7507 and 0.90, suggesting its relevance in aging.
