SIRT1 Controls the Final Step of Cell Recycling and Mitochondria Cleanup in Heart Muscle Cells

Updated

Abstract

SIRT1 is identified as a regulator of autophagosome-lysosome fusion, promoting autophagosome degradation.

  • Mitochondrial stress from CCCP decreased mitochondrial protein levels and increased phosphorylation of ubiquitin in cardiomyocytes.
  • Co-treatment with chloroquine prevented CCCP-induced decreases in mitochondrial proteins, indicating the activation of mitophagy.
  • SIRT1 knockdown led to the accumulation of fragmented mitochondria and autophagosomes without affecting ubiquitin phosphorylation.
  • Impaired autophagosome-lysosome fusion was observed following SIRT1 knockdown, as demonstrated by colocalization analyses.
  • In vivo, cardiomyocyte-specific SIRT1 knockout mice showed elevated LC3-II levels and reduced autophagic flux.
  • SIRT1 deficiency reduced autophagosome degradation in a model treated with doxorubicin.
