Abstract

F1462, a novel SIRT1 up-regulator, significantly reduces markers of vascular endothelial cell senescence and inflammation.

  • F1462 decreased the levels of senescence markers p53 and p21 in a dose-dependent manner.
  • The compound lowered inflammatory factors associated with cellular senescence.
  • F1462 improved mitochondrial function by restoring mitochondrial membrane potential and reducing reactive oxygen species generation.
  • In naturally aged mice, F1462 reduced plasma inflammatory factors linked to aging and decreased vascular wall thickening.
  • Activation of the SIRT1/PGC-1α/TFAM signaling pathway was identified as a mechanism for the benefits of F1462.
