Single-cell transcriptome analysis of aging ovaries revealed elevated mitochondrial metabolism and disrupted histone modifications.

  • Aging ovaries exhibited hallmark alterations associated with cellular senescence.
  • Impaired SIRT5-mediated desuccinylation was identified as a key driver of ovarian aging.
  • SUCLG2, a protein in the TCA cycle, is the main target of SIRT5-mediated desuccinylation.
  • Desuccinylation of SUCLG2 improved mitochondrial function during cellular senescence.
  • Oversupply of acetyl-CoA accelerated ovarian aging by increasing specific histone acetylation.
  • Gene therapy targeting SUCLG2 desuccinylation mutants showed potential to alleviate ovarian aging.
