Aging

Small cell-released particles may carry aging signals to nearby cells in lab models

Updated

Abstract

Fibroblasts expressing p16INK4A release significantly more small extracellular vesicles (sEVs) than proliferating cells.

  • Small extracellular vesicles from p16INK4A-expressing fibroblasts are biologically active and can induce senescence in other cells.
  • Treatment with these sEVs leads to proliferative arrest, increased p16INK4A expression, and activation of a DNA damage response in recipient cells.
  • sEVs from p16INK4A-expressing fibroblasts do not stimulate the production of the typical inflammatory factors IL-6 and IL-8.
  • The transmission of paracrine senescence via sEVs is observed in models of aging, including progerin expression and telomere dysfunction.
  • sEVs from fibroblasts of aged donors can induce paracrine senescence in younger fibroblasts.
