Targeted Treatment of Liver Cancer Using Calcium Phosphate Lipid Nanoparticles Carrying Bcl-2 siRNA and Doxorubicin Modified with SP94

Updated

Abstract

SP94-LCP/DOX&Bcl-2 siRNA demonstrated a mean diameter of 130 nm with a 92% encapsulation efficiency for siRNA.

  • The formulation showed a zeta potential of 20 mV and a spherical morphology with uniform distribution.
  • A significant sustained-release effect was observed, indicating good stability of the nanoparticles.
  • SP94-LCP/DOX&Bcl-2 siRNA significantly lowered the IC₅₀ values for doxorubicin in combination with Bcl-2 siRNA.
  • Enhanced cellular uptake of HepG2 cells for the nanoparticles was noted, along with a remarkable inhibition of cell migration.
  • The treatment resulted in downregulation of Bcl-2 and upregulation of Bax, promoting apoptotic cell death.
  • In vivo, the formulation effectively inhibited tumor growth in mice while reducing toxicity to normal tissues.
