Creating human stem cells with spCAS9 to study gene roles in bone cell development

Abstract

A stable CAS9 expressing human bone marrow-derived stromal cell line (hMSC-TERTCAS9) has been engineered to facilitate efficient gene editing.

  • This cell line allows for efficient and reproducible gene editing through the constitutive expression of spCas9.
  • Edited cells can be sorted and expanded to create homogenous clonal cell lines with mono- or bi-allelic gene deletions.
  • The hMSC-TERTCAS9 cells have been validated for studying gene function during cell proliferation and differentiation.
  • Indel generation and clonal selection were used to inactivate the vitamin D receptor and create larger gene disruptions.
  • Precise removals of small genomic sequences have also been achieved, demonstrating the versatility of this cell line.
