Autophagy is a critical cellular mechanism that regulates the degradation of misfolded proteins and non-functional organelles.

  • Autophagy can be classified into three major types: macroautophagy, microautophagy, and chaperone-mediated autophagy.
  • Sequestosome 1 (SQSTM1)/p62 functions as a selective autophagy receptor that binds misfolded proteins for degradation.
  • p62 integrates multiple signaling pathways, including those involving NF-κB, mTORC1, and NRF2.
  • Phase separation of p62 is an important regulatory process within the autophagy mechanism.
  • Impaired or excessive autophagy mediated by p62 is associated with cancer development.
