Autophagy

Reducing SSH1 boosts cell cleanup by attracting ATG9A and helps break down tau protein

Updated

Abstract

Reduction of the phosphatase SSH1 consistently lowered steady-state MAPT/tau levels by enhancing autophagic flux.

  • Modulation of specific genes mitigated MAPT/tau-induced behavioral deficits in Drosophila models.
  • Depletion of SSH1 promotes mobilization of ATG9A to the endoplasmic reticulum-phagophore assembly site, accelerating autophagosome formation.
  • Inhibition of SSH1 enhances autophagic degradation of MAPT/tau through a mechanism distinct from its regulation of CFL1.
  • SSH1-mediated clearance of MAPT/tau requires functional ATG9, as shown by the effects in Drosophila models.
  • Experiments indicated that SSH1 decreases ATG9A phosphorylation in the presence of actin, suggesting a regulatory interaction.
