Staphylococcus aureus changes arginine use to promote mammary gland aging through immune and tissue cell interaction

Abstract

Localized tissue senescence was observed in the mammary glands of mice with chronic S. aureus infection.

  • Chronic S. aureus infections are linked to significant tissue and organ dysfunction due to difficulty in elimination.
  • In a mouse model, S. aureus infection disrupted the ornithine cycle in mammary tissues.
  • Arginase 1 (Arg1) was identified as a key regulator in the metabolic changes following infection.
  • Stimulation by S. aureus caused mammary epithelial cells to produce cytokines, leading to increased Arg1 release from macrophages.
  • Elevated Arg1 in mammary tissue promotes the conversion of arginine to ornithine, contributing to cellular senescence.
  • Supplementing with excess arginine or ornithine can replicate the senescence phenotype, while reducing Arg1 can reverse it.
