STX4 is essential for maintaining healthy energy centers in skeletal muscle

Abstract

Over 50% reduction of Syntaxin 4 (STX4) in skeletal muscle is associated with insulin resistance in male mice.

  • Knockout of STX4 in male mice led to decreased energy expenditure and respiratory exchange ratio.
  • Impaired mitochondrial oxygen consumption rate was observed in STX4-deficient muscle.
  • Mitochondrial morphology was altered, with fragmented mitochondria in oxidative muscle and swollen mitochondria in glycolytic fibers.
  • Reduced mitochondrial DNA levels and expression of mitochondrial biogenesis genes were noted in STX4-depleted muscle and myotubes.
  • Impaired mitochondrial turnover and decreased levels of mitophagy markers were observed in both STX4-deficient muscle and myotubes.
