Sunitinib may cause removal of aging cells by changing a growth factor receptor

Updated

Abstract

Sunitinib reduced senescent cells and suppressed SASP expression in a mouse model of lung fibrosis.

  • Receptor tyrosine kinases (RTKs) play a key role in regulating cell survival and responses to environmental changes.
  • Sunitinib, a multi-targeted RTK inhibitor, selectively induced cell death in senescent human dermal fibroblasts by activating specific proteins involved in apoptosis.
  • Increased PDGFRβ phosphorylation was observed in aged cells, suggesting its role in senescence.
  • Knockdown of PDGFRβ made senescent cells more prone to death without affecting non-senescent cells.
  • The JAK2-STAT3 pathway was implicated in the senolytic effects of sunitinib, while JAK1 phosphorylation was associated with resistance in non-senescent cells.
