Survivin mRNA lipid nanoparticles activate killer T cells and memory response for effective oral cancer immunotherapy

Updated

Abstract

Complete tumor elimination was achieved within 7-10 days after administration of survivin mRNA lipid nanoparticles in a murine oral cancer model.

  • Survivin mRNA lipid nanoparticles induced significant immune reprogramming, leading to enhanced activation of immune cells, including dendritic cells and T cells.
  • A broad, multi-compartmental T-cell immune response was observed following survivin mRNA-LNP treatment compared to control.
  • Significantly greater expansion of CD4+ and CD8+ T cells was noted in lymph nodes by day 3 and robust infiltration into the tumor microenvironment by day 14.
  • Survivin mRNA-LNPs promoted T cell activation and increased cytotoxic CD8+Granzyme B+ T cells in the tumor microenvironment.
  • Controlled Treg levels were maintained, resulting in a favorable effector to Treg ratio that is critical for effective anti-tumor responses.
  • Tumor rechallenge showed the development of immunological memory against oral cancer cells.
