Improved sustainable sugar-based gene-delivery systems using free polyethyleneimine chains

Abstract

Hybrid gene-delivery platforms based on permanently cationic polysaccharides were developed to enhance transfection efficiency while maintaining cell viability.

  • Permanently positive charges on dextran and chitosan backbones enable strong binding with DNA and formation of polymer-DNA complexes.
  • Grafted poly(2-oxazoline) segments improve the stability of these carriers in serum.
  • Incorporating small amounts of free branched polyethyleneimine (BPEI) significantly enhances transfection efficiency without compromising cell viability.
  • Free BPEI chains may assist in promoting endosomal escape and the release of DNA inside cells, but are not necessary for initial DNA condensation.
  • The formulation strategy is modular and sustainable, enabling high gene expression with reduced toxicity, making it suitable for gene therapy applications.
