Reducing harmful molecules in joint tissue may lower cell death and slow osteoarthritis

Updated

Abstract

Post-synthetic Mn incorporation enhanced the superoxide dismutase and catalase-like activities of GaMOF.

  • Mn@GaMOF regulates the synovial nitro-oxidative microenvironment by targeting multiple reactive species.
  • The framework reduces the accumulation of reactive oxygen species, nitric oxide, and reactive nitrogen species.
  • In macrophages, Mn@GaMOF decreases inflammatory cytokine production and modifies inflammatory marker expression.
  • Mn@GaMOF mitigates nitro-oxidative stress and inflammatory activation in fibroblast-like synoviocytes.
  • In a mouse model of osteoarthritis, intra-articular administration of Mn@GaMOF alleviated pain and synovial inflammation.
  • This approach may represent a relevant therapeutic strategy for managing osteoarthritis by targeting the interconnected redox network.
