Targeted removal of sugar-damaged cell parts reverses diabetic heart disease in mice and lab-grown human heart tissue

Updated

Abstract

Glycophagy dysregulation is associated with impaired diastolic relaxation in diabetic heart disease.

  • Diabetic heart disease is characterized by early impaired diastolic relaxation.
  • Glycogen accumulation and disrupted glycogen autophagy have been found in the diabetic heart.
  • The glycogen receptor protein STBD1 is mobilized early in response to metabolic challenges.
  • Deficiency of the ATG8 family protein GABARAPL1 is linked to diastolic dysfunction in diabetes.
  • Gene delivery of GABARAPL1 improved cardiac diastolic function in type 2 diabetic mice.
  • Diastolic performance was enhanced in human stem cell-derived cardiac organoids with GABARAPL1 treatment.
