Abstract

Targeted lipid nanoparticles can enhance transfection efficiency significantly, achieving up to 20 times greater liver transfection compared to nontargeted nanoparticles.

  • Targeted lipid nanoparticles (tLNPs) enable receptor-mediated uptake in difficult-to-transfect cells.
  • Conjugation of antibodies to LNPs can reduce the required dosage for effective protein expression.
  • In vivo studies show that antibody conjugation enhances targeting efficiency in specific tissues, with a fivefold increase in lung transfection and a twentyfold increase in liver transfection.
  • Ex vivo targeting to primary human T cells shows a 4.5 times increase in uptake and significant enhancement of mRNA transfection.
  • The described modular strategy allows for compatibility with various LNP formulations or antibodies.
