Abstract

Essence

A PEG-free HER2-targeted mRNA-LNP platform showed tumor-directed delivery and antitumor activity in preclinical models.

Evidence

This formulation/platform experiment assembled palmitoylated nanobody, lipids, and spike-protein mRNA into targeted LNPs, then tested physicochemical properties, in vitro tumor-cell killing, tumor delivery and retention, and in vivo immune killing with tumor volume reduction.

Caveat

The evidence is preclinical and relies on spike-protein expression and pre-existing immunity, so therapeutic performance in human cancers remains untested.
