How Telomerase Location Inside Neurons Affects Control of Mitochondrial DNA Amount in Specific Genes

Abstract

Mitochondrial-targeted TERT (mito-TERT) upregulated the copy number of mitochondrial complex I genes in neuronal cells.

  • Oxygen-glucose deprivation/reoxygenation caused TERT to redistribute from mitochondria to the nucleus in neuronal cells.
  • Both mitochondrial-targeted (mito-TERT) and nuclear-targeted (nuc-TERT) versions of TERT were successfully created.
  • Mito-TERT specifically increased the copy number of mitochondrial complex I genes, while nuc-TERT upregulated a broader range of mitochondrial genes.
  • Mito-TERT was found to bind directly to the promoters of the ND1 and ND2 genes.
  • The findings suggest that TERT regulates mitochondrial DNA copy number through distinct mechanisms based on its location within the cell.
