Using molecules to change special DNA structures and their effects on telomere maintenance by DNA:RNA hybrids and alternative lengthening methods

Updated

Abstract

Essence

New TERRA ligands increased telomeric DNA:RNA hybrids and -related features in cancer cells.

Evidence

A ligand-based virtual screen of FDA-approved drugs followed by cell and in vitro experiments in telomerase-positive and ALT-positive cancer cells identified TERRA G-quadruplex ligands that stabilized TERRA-chromatin binding, increased telomeric DNA:RNA hybrids, telomeric defects, ALT-associated PML bodies, and partly C-circles in an RNAseH1-dependent manner.

Caveat

The findings are mechanistic cell and in vitro results, and some ALT readouts increased only partly rather than showing direct therapeutic effects.

What this is

  • The research investigates the role of TERRA G-quadruplexes (GQs) in telomere dynamics and cancer biology.
  • TERRA, a long non-coding RNA, is involved in telomere protection and is overexpressed in -positive cancer cells.
  • The study identifies FDA-approved drugs that stabilize TERRA GQs, impacting telomeric DNA:RNA hybrids and mechanisms.

Essence

  • FDA-approved drugs that stabilize TERRA G-quadruplexes increase telomeric DNA:RNA hybrids and induce telomeric defects, enhancing activity in cancer cells.

Key takeaways

  • TERRA stabilization by specific FDA-approved drugs leads to increased formation of DNA:RNA hybrids at telomeres, contributing to telomeric defects.
  • Treatment with ligands fedratinib, netarsudil, and osimertinib significantly increases the mean number of -associated PML bodies in cancer cells, indicating enhanced activity.
  • The study suggests that TERRA ligands can induce telomeric abnormalities characteristic of -positive cells, potentially influencing cancer progression.

Caveats

  • The study primarily focuses on TERRA stabilization and its effects, but other mechanisms of action for the drugs may also play a role.
  • The findings may not fully account for the complex interactions between TERRA, GQs, and other cellular processes affecting telomere dynamics.

Definitions

  • G-quadruplex (GQ): A four-stranded nucleic acid structure formed by guanine-rich sequences, stabilized by Hoogsteen hydrogen bonds.
  • ALT (alternative lengthening of telomeres): A telomere maintenance mechanism that does not involve telomerase, relying on homologous recombination.
