TGR5 helps prevent learning and memory problems from long-term stress in mice by controlling gut-related inflammation

Abstract

Chronic restraint stress (CS) for 21 days led to significant impairments in memory and anxiety-like behaviors in mice.

  • CS resulted in decreased body weight gain and triggered anxiety-like behaviors in the tested mice.
  • Significant damage was observed in the hippocampal CA1 region and colon, with increased TGR5 expression in both tissues.
  • CS altered the composition of gut microbiota and bile acid metabolism, particularly increasing TCDCA levels, which may contribute to neuroinflammation.
  • TGR5 deficiency exacerbated CS-induced neuroinflammation, as indicated by elevated levels of pro-inflammatory markers, including IL-1β and TNF-α.
  • CS decreased serotonin levels and impaired long-term potentiation, disrupting synaptic plasticity and contributing to learning and memory deficits.
  • In the colon, TGR5 deficiency also worsened tissue injury associated with chronic stress.
