Rapamycin reduces plaque inflammation and immune activity in old mice with artery disease

Abstract

Rapamycin treatment reduced plaque macrophage content to 0.73 ± 0.07 × 10⁵ μm² in aged atherosclerotic mice.

  • Treatment with rapamycin decreased total T cell numbers in both vascular and lymphoid tissues.
  • There was a shift from effector T cell types to central memory phenotypes, especially in CD8+ T cells.
  • Regulatory CD4+ T cells showed an increase in number and expression of suppressive genes after rapamycin treatment.
  • Rapamycin led to a reduction in T follicular helper cells and associated immunoglobulin levels.
  • Age-associated B cells were diminished in both spleen and lymph nodes, indicating an anti-inflammatory effect.
  • Aortic leukocytes exhibited significantly reduced markers of cellular aging.
