Immune-Boosting Effects of Human IFN-β mRNA in Primary Human Myeloid Cells Depend on Timing

Updated

Abstract

IFN-β mRNA induced time-dependent apoptosis, most pronounced in HepG2 cells.

  • IFN-β mRNA triggered CXCL10 secretion in both cell lines tested.
  • In primary immune cells, IFN-β mRNA activated dendritic cells and increased pro-inflammatory cytokine secretion.
  • M2-polarized macrophages were repolarized toward a pro-inflammatory phenotype by IFN-β mRNA.
  • During co-transfection, IFN-β mRNA enhanced dendritic cell activation, but antigen expression was maintained only when antigen mRNA was delivered before IFN-β mRNA.
