Improved Weight Loss by Keeping Tirzepatide in Fat Tissue Using a Special Coating That Boosts Leptin Receptor Signals

Abstract

Localized injection of a TZP encapsulated formulation increased weight loss by dramatically enhancing retention in inguinal white adipose tissue (iWAT) compared to TZP alone.

  • Retention of TZP in iWAT was significantly improved by encapsulation in mesoporous polydopamine.
  • DIO mice receiving the encapsulated TZP showed greater weight loss and better plasma lipid profiles than those receiving TZP alone.
  • The encapsulated TZP injection improved key metabolic processes, such as leptin resistance and lipid metabolism, in iWAT.
  • Leptin receptor deficiency in db/db mice negated the weight loss benefits of the encapsulated TZP injection, although glucose-lowering effects remained consistent.
  • These results suggest a tissue-specific action of TZP that could inform targeted delivery strategies for obesity treatment.
