How Well Different Doses of Tirzepatide and Semaglutide Help Weight Loss and Are Tolerated in Adults with Obesity Without Diabetes

Abstract

Tirzepatide at maximum tolerated dose (MTD) resulted in a 20.90% mean difference in weight loss compared to placebo.

  • All active treatments led to significantly greater weight loss compared to placebo.
  • Tirzepatide MTD showed the highest efficacy for weight loss among the studied doses.
  • Similar dose-dependent reductions were observed for waist circumference and achieving ≥15% weight loss.
  • The evidence for achieving ≥15% weight loss was high for most interventions, but moderate for semaglutide MTD and tirzepatide 5 mg.
  • Semaglutide MTD had the highest likelihood of treatment discontinuation due to adverse events.
  • Gastrointestinal side effects were more prevalent with higher doses of both tirzepatide and semaglutide.
