TLR4 mutation lowers brain immune cell response, raises amyloid buildup, and worsens memory loss in an Alzheimer's mouse model

Abstract

At 5 months, there was no difference in cerebral Aβ load between AD mouse models with and without TLR4 mutation.

  • Microglial activation was less in TLR4 mutant AD mice compared to wild-type AD mice at 5 months.
  • At 9 months, TLR4 mutant mice showed increased Aβ deposition and soluble Aβ42 in the brain.
  • Decrements in cognitive functions were observed in TLR4 mutant mice compared to wild-type mice.
  • Expression levels of IL-1β, CCL3, and CCL4 in the hippocampus were lower in TLR4 mutant mice at 9 months.
  • TLR4 mutation reduced Aβ-induced expression of IL-1β, CCL3, and CCL4 in monocytes.
