Abstract

The engineered platform showed effective tumor regression in colorectal cancer through a multifaceted immunotherapy approach.

  • A tumor microenvironment-responsive platform was developed using polyethylene glycol-coated solid lipid nanoparticles for targeted therapy.
  • The platform co-delivered a VCP/p97 inhibitor and bispecific aptamers to induce endoplasmic reticulum stress and block immune checkpoints.
  • Induction of endoplasmic reticulum stress led to immunogenic cell death and apoptosis, while also suppressing cancer stem cell-like characteristics.
  • The treatment reactivated T cells and enhanced levels of proinflammatory cytokines while suppressing immunosuppressive factors.
  • In vivo imaging confirmed selective accumulation of the treatment in tumors and demonstrated effective tumor regression.
