Tomatine boosts mRNA vaccine responses as a flexible helper

Abstract

Tomatine-based liposomes (T-LPX) significantly enhanced mRNA expression efficiency and immune activation.

  • Tomatine improved uptake of mRNA in dendritic cells through a specific cellular process.
  • Increased expression of a key protein was linked to enhanced mRNA translation.
  • Tomatine activated a specific immune pathway, leading to increased secretion of immune signaling molecules.
  • Co-stimulatory molecules in dendritic cells were upregulated following tomatine treatment.
  • T-LPX loaded with tumor antigen-encoding mRNA induced strong immune responses in a mouse tumor model.
  • Tomatine's adjuvant effects were also effective with different types of nanocarriers and antigens.
