Traditional Chinese Herbal Formula YA3D3 Improves Alzheimer’s-Like Brain Changes and Memory Problems in a Mouse Model

Abstract

YA3D3-MHF demonstrated stronger antioxidant activity than YA3D3-HF in vitro (P<0.001).

  • Network pharmacology highlighted significant involvement of YA3D3 targets in autophagy and related signaling pathways.
  • Both YA3D3 preparations reversed the decrease in ATG14 and ATG16L1 expression caused by ATG14 knock-down in BV2 cells, with YA3D3-MHF showing greater potency (P=0.003).
  • Mice treated with YA3D3 exhibited shorter escape latencies and more platform crossings in the Morris water maze compared to the APP/PS1 model group (all P<0.05).
  • Cerebral Aβ fluorescence intensity was reduced in all YA3D3 groups (all P<0.001).
  • YA3D3 up-regulated ATG14 expression and modulated downstream autophagy-related gene expression in brain tissue (all P<0.05).
