Gene activity linked to antidepressant resistance in brain emotion and thinking areas of chronically stressed mice

Abstract

Approximately one-third of individuals with major depressive disorder are affected by treatment-resistant depression (TRD).

  • A preclinical mouse model of TRD was developed using chronic social defeat stress and treatments with fluoxetine (FLX) and ketamine (KET).
  • RNA sequencing of brain regions revealed specific gene expression patterns linked to treatment responses in mice.
  • Previous treatment with FLX enhanced the effectiveness of KET in some mice, while non-responsive mice showed no such benefit.
  • Distinct transcriptional profiles were observed in the nucleus accumbens and prefrontal cortex of responders versus non-responders.
  • Gene analysis identified unique gene modules associated with stress susceptibility and antidepressant resistance, indicating different underlying mechanisms.
