Thirty-one transcriptional regulators of microglial activation states were identified through CRISPR interference screens.

  • Microglia can exhibit diverse activation states that are important for brain health and disease.
  • Dysregulated microglial activation is associated with various brain disorders.
  • Inhibiting specific regulators may affect microglial functions like interferon response and lysosomal activity.
  • Certain knockdowns led to increased disease-associated signatures and enhanced phagocytosis.
  • Widespread loss of methylation was observed with specific knockdowns, which may activate negative regulators of interferon signaling.
