A modified chalcone compound reduces Alzheimer's Aβ and Tau problems by affecting energy control and cell cleanup processes

Updated

Abstract

Compound 51 (C51) improved learning and memory in 3×Tg-AD mice while reducing Aβ oligomer accumulation and phosphorylated Tau burden.

  • C51 enhanced autophagic processes, lysosomal activity, and turnover of SQSTM1/p62 in mammalian cells and Caenorhabditis elegans.
  • C51 mitigated the toxic effects associated with APP, Tau, and TauP301L in neuronal cells and improved behavioral deficits related to Aβ and Tau in transgenic C. elegans.
  • In 3×Tg-AD mice, C51 preserved hippocampal neurons and synaptic proteins, and reduced gliosis without causing systemic toxicity.
  • C51 activated AMP-activated protein kinase (AMPK) and inhibited mechanistic target of rapamycin (mTOR) signaling, indicating a mechanism for its effects.
  • AMPK signaling was necessary for C51's protective effects against Aβ/Tau toxicity, as shown by experiments involving AMPK knockdown and RNA interference.
